Olive oil is one of the most studied foods in nutrition science, with more than 2,000 papers on its polyphenols alone. These are the compounds that make fresh olive oil bitter and peppery: hydroxytyrosol, oleocanthal, oleacein and their relatives. They are also what researchers measure when they test olive oil in people.

One pattern keeps coming back in that research. The oils with more polyphenols are the oils that show more.

What are olive oil polyphenols?

Olive oil polyphenols are natural compounds in extra virgin olive oil, including hydroxytyrosol, oleocanthal and oleacein. They give fresh olive oil its bitterness and peppery sting, and they work together with other compounds in the oil, such as squalene and vitamin E.

What is oleocanthal and why does it matter?

Oleocanthal is the olive oil polyphenol that causes the peppery sting at the back of the throat. In laboratory tests it acts on the COX enzymes involved in inflammation, and in a randomised trial in which oils were matched for total polyphenols, the effect on blood platelets tracked oleocanthal content (Agrawal et al., 2017). Most olive oils contain little of it.

What does research show on olive oil and the brain?

In a randomised trial in people with mild cognitive impairment, high-phenolic extra virgin olive oil reduced blood-brain barrier permeability in memory regions of the brain, while refined olive oil did not (Kaddoumi et al., Nutrients, 2022).

How do you take Oleaphen?

One 5 ml monodose a day, on its own or over food once it is off the heat.

​Does polyphenol concentration matter?

Yes. In a 2025 randomised trial published in Nutrients, two groups received the same daily amount of olive oil polyphenols; the group taking a smaller serving of a more concentrated oil (1,021 mg/kg) had a greater fall in total cholesterol than the group taking a larger serving of a 414 mg/kg oil.

How much oleocanthal does Oleaphen contain?

Oleaphen's 2025 harvest contained 1,248 mg/kg of oleocanthal within 2,236 mg/kg of total polyphenols, measured by LC-MS/MS at Universidad de Córdoba. One 5 ml daily dose carries about 5.7 mg of oleocanthal, as much as more than a cup (about 270 ml) of extra virgin olive oil at the 23 mg/kg measured in US oils. Certified 2026 results are pending, and first lab results are higher than 2025.

​Is there an approved health claim for olive oil polyphenols?

​Yes. In the EU, olive oil polyphenols contribute to the protection of blood lipids from oxidative stress, for an oil providing at least 5 mg of hydroxytyrosol and its derivatives per 20 g, with a daily intake of 20 g (EU Regulation 432/2012). In the US, the FDA allows a qualified health claim for olive oil: limited and not conclusive scientific evidence suggests that eating about 2 tablespoons (23 grams) of olive oil daily may reduce the risk of coronary heart disease due to the monounsaturated fat in olive oil. To achieve this possible benefit, olive oil is to replace a similar amount of saturated fat and not increase the total number of calories you eat in a day.
